Paclitaxel was added to one of the culture dishes at a concentration of 100nM.to the third culture dish, cytochalasin D was added at a concentration of 100nM Upon the establishment of the three treatment conditions, the culture dishes were then incubated for 48 hours to give time of the re-epithelialization to take place. After incubation, each of the culture dishes was fixed using 10% neutral and buffered formalin for preservation purposes. Each of the fixed culture dishes was stained by use of eosin and hematoxylin stains. Re-epithelialization of the wound was monitored over time by observing the migration of the cells. This was measured by measuring the change in the thickness of the wound from the time a wound was introduced to the ultimate healing. The total change in the thickness of the wound was calculated against time. The organization of the microfilaments and was also determined by exposing the cells to varied conditions. To achieve this, the f-actin was labelled fluorosly by use of FITC- conjugated phalloidin. The network of the microfilaments was then observed by use of the fluorescence microscope. It was observed that as time passed by, the epithelial cells within the margin of the wound migrated into the area of the wound. This was in an effort to replace the cells that had been scratched away as a result of tissue injury. Moreover, it was observed that the cells that remained in the margin of the wound underwent increased proliferation to replace the cells that had migrated to the wounded area of the tissues. With time, the thickness of the injured area decreased. The addition of dimethyl sulfoxide as a control in this assay was in order to account for the possibility of any nonspecific effect of this solution on the wound healing. Paclitaxel was added to a different culture dish in order to stabilize the microtubules in the cells by interfering with the division and migration of cells (Cai et al., 2014). The addition of cytochalasin D was in order to find out the roles played by microfilaments in healing of wounds. This is because cytochalasin D blocks the reorganization of microfilaments and hence it affects proliferation and cell migration in a negative way (Kamimura et al., 2015). This experiment or assay clearly gives an indication of the way in which the cells behave upon injury. The healing of the wound occurred in a stereotyped way, that is, the cells polarized in a negative way to the wound area. The polarized cells then initiated the protrusion and migration and finally the wound became closed. The time based microscopy provides an e fficient tool via which the events of cell migration and healing of the wound occurs in a sequential manner. Some of the practical examples of this assay are the proliferation of cells and deposition of the extracellular matrix, and an increase in inflammatory cells. In another approach, this assay is important in explaining the way in which a disease is distributed in the body from an initial infected organ through metastasis. The PC-3 cells were stained in order to increase the contrast during observation under the fluorescent microscope.it is therefore important to consider the methods of staining because some methods can damage the cells being studied. Some staining methods require the dyes to penetrate deep into the cell, something which is not possible in live cells. In order to curb this and make it important to store the stained cells for future reference, fixation, which involves killing of the cells while maintaining their structure and composition is carried out before st aining (Vasicova et al., 2016). Fixation can be done by use of the chemicals like the buffered solution containing formaldehyde. This chemical is known to maintain the integrity and structure of organelles and structure of the cells while at the same time blocking the decomposition of cells by enzymes. Since this chemical is carcinogenic, it needs to be handled with great caution (Kuzmin et al., 2014). The physical methods of fixation involves either drying of freezing of cells. The choice of the fixation methods to be used depends on a number of factors such as types of cells being used and the intention of the experimental set up. In most cases however, the formaldehyde is used for fixation purposes. The preservation of cells using this chemical is made possible through a process which forms cross bridges between the nucleic acids and the amines on the proteins (Andreeva et al., 2016). In my essay about cafeteria food, I will explain what students want in their food. I will also share the likes and dislikes in the food and what we can do to improve it. Cold pizza, undercooked hamburgers and brown lettuce sounds nasty. Many students from different states and districts have to deal with that every day at their schools. The food from the cafeterias used to be good, with a sweet flavor, but in the last few years the food had taken a different flavor, a flavor that cannot be tolerated by the students. The students need a better quality in their food, don’t they? It’s time for a change, a change to food, so students can be satisfied while their eating, a change to food that can be really worth it. As a matter of fact, we students pay money to the district every year and this is what we get? It’s time for a change. Everybody knows that eating healthy was from the past. However, now students and not necessarily just students eat fast food like McDonalds or Taco Bell. Now everybody does, which can be a good and a bad thing. The changes of the food’s taste has made many students to not eat at school, or even made students to bring lunch, and this as a matter of fact causes the school to lose money. In all actuality is the loss of money causing the school to give students poorly made food? If the schools would serve students food from places like Pizza Hut and Subway I can guarantee that most or all students would eat at the cafeterias and it would benefit to both school and students. These types of foods would be so great to have in schools, but unfortunately the district and the governments have taken action and unfairly, they have set down laws that prohibit these foods in the schools. They have settled these laws because they want us as students to eat healthy, but if they want to keep us healthy, why are they giving us food with a bad and nasty taste? Also, with these laws they keep students like me away from eating too much â€Å"junk† food because they can cause obesity. But do they keep on giving us food that is not even worth the amount of money our parents give to the government every year? If they were the ones who eat all of the nasty food given by the cafeteria they would of get tired of eating cold pizza and uncooked hamburgers with brown lettuce just like we the students do. In conclusion, we all understand what the government and the school system is trying to do with this healthy eating program. They want to promote healthy eating habits in an effort to prevent obesity. Although nutrition is important, students feel that there is a better way to have healthy food without ruining the taste. Therefore, in an effort to keep students healthy and leave with their stomachs satisfied we should all come together to realize that we are the ones that have to make it through the school day with the food. Cover Girl Advertisement

When thinking of advertisements, for some odd reason, I immediately will think of Cover Girl makeup ads that are often found in almost every woman’s magazine. However, this particular advertisement features country music superstar Taylor Swift and very similar to all of their distinctive ads focuses mostly on her face. This ad is for their new â€Å"natureluxe silk foundation- luxury touched by nature. † From this relatively simple ad, as it only contains a close up of Taylor Swift as well as text, it allows for the discussion of it’s unique aesthetics. The first thing that I would like to discuss is the colors that are used within this advertisement. The advertisement finds balance by using a soft and light yet trendy green for their top lettering of Cover Girl, some pop out lettering under the image all to match the color of the actual foundation bottle. The advertisement becomes more appealing to the eye as Cover Girl is attempting to create an earthy yet â€Å"simple is beautiful† feel for their audience. Also, the actual colors of Taylor Swift herself are very fair and soft as it looks as if she is not wearing any foundations. In addition, she is wearing a pale pink ruffled dress adding to the light, fluffy and earthy feel of the advertisement as a whole. However, the majority of the wording is a dark black with some of the font in bold and different sizes. It is evident that the advertisers are attempting to bring their audience’s attention down from Taylor Swift to what they actually have to say about the product by using opposing colors and bold text. In addition to the colors that were used, the actual wording that was used is particularly interesting. The advertisement uses many different sizes and colors within the whole image. The top Cover Girl is in the light green color reflecting back to the idea of softness and it is the vantage point for a person’s eyes. It is very bold and automatically draws attention to the top half of the advertisement. Also, the different very black and dark bold text at the bottom of the page seems intriguing for a few reasons. First, the different font types and sizes attempt to make you really see the first three lines very clearly and get an understanding of what the product is. However, the work â€Å"new† happens to be in green followed by â€Å"natureluxe silk foundation† with â€Å"luxe† being bolded, suggesting that this product is truly â€Å"luxury touched by nature;† which is in very small print right below that as well. In addition, I find it interesting that the two black bolded lines both have the word â€Å"air† in them and also soft, air-like words, such as â€Å"fresh† and â€Å"breath. To me this is suggesting that even though the text may be dark and bold, this product is still going to make the user feel fresh and lightweight. Therefore, this brings me to the last topic of the lighting that is used within the image as well as the text. With the text having a dark and bold at the top, it slowly becomes lighter and smaller and even throws in come light green text in there as well, suggesting that it almost has a elegant, luxurious feel to the text just as much as the makeup is going to have on a person’s face. Also, the lighting within the image is very bright and the backdrop of a complete white again is keeping the theme of lightweight and airy flowing throughout the entire advertisement. In addition, the bright, artificial white lighting is also used on Taylor Swift’s face as well as arm to suggest that she is feeling fresh and clean when wearing this foundation.
